Over the years, we have extensively utilized ddPCR for a variety of gene copy number tests, particularly in support of gene-edited cell therapies, including CNV analysis and translocation studies.

- CAR-T CNV Analysis: We use validated dual TaqMan™ assays and ddPCR to determine lentiviral or retroviral gene copy number, normalized to endogenous genes, offering superior accuracy over traditional qPCR.
- DNA/RNA Quantitation: ddPCR enables highly sensitive quantification of gene and viral titers in preclinical and clinical samples, eliminating recovery loss from extraction while maintaining consistent precision.
- Translocation Studies: For gene editing safety assessments, ddPCR detects rare translocation events with customizable long amplicon designs, improving sensitivity compared to qPCR.
- CGT PK Studies: ddPCR supports pharmacokinetic evaluations of therapeutic cells and vectors in animal or human studies by pooling reactions to increase assay volume and sensitivity.
- Gene Expression Analysis: ddPCR provides a robust solution for tracking therapeutic gene expression, enabling high-sensitivity pharmacodynamic studies with pooled sample reactions.
- Rare Mutation Detection: ddPCR delivers unmatched sensitivity for identifying low-frequency mutations in cancer or CGT vectors, outperforming qPCR in detecting minor genetically modified populations.
