Client Challenge

A cell therapy developer needed a functional bioassay to demonstrate the anti-inflammatory activity of its drug product, first to characterize the assay’s operating parameters and then to validate it for use as a cGMP quality control test. The assay needed to show that the drug product suppressed a defined inflammatory response in a standard immune cell model, with specificity, accuracy, and precision that held up across operators and days.

Avance Biosciences’ Solution

Avance Biosciences pre-validated and then validated a cell-based anti-inflammatory functional assay. The method:

  • Stimulates a macrophage reporter cell line with a standard pro-inflammatory stimulus to trigger a reproducible inflammatory response
  • Treats the stimulated cells with the drug product, or with a small-molecule reference compound as a positive control for inhibition
  • Measures a pro-inflammatory cytokine in the treated-cell supernatant by ELISA as the inflammatory readout
  • Calculates percent inhibition of the cytokine relative to stimulated, untreated cells as the anti-inflammatory activity measure

Project Overview

The assay was first run as a pre-validation study, performed by three operators, to confirm the cell-based and ELISA steps worked as designed, establish acceptance criteria, and identify a reliable working range for the detection ELISA. The confirmed method then moved into a formal validation study, executed across three operators, three separate days, and nine independent assay runs.

Assay Validation

Each parameter answered a question a quality unit will ask of a potency method:

  • Specificity: Does the assay respond only to the intended stimulus? Unstimulated controls, stimulated samples, and the ELISA’s own internal negative control all measured within their predefined ranges on every run, and the positive control met its predefined inhibition criterion.
  • Accuracy: Does the ELISA return the right concentration? Percent recovery across the validated quality control panel met the predefined acceptance criterion on every run.
  • Precision: Does the result hold up across replicates, operators, and days? Intra-assay, inter-day, inter-operator, and intermediate precision were each evaluated across the nine runs, and variability in both the drug product’s and the positive control’s inhibition readouts, and in the detection ELISA’s quality control samples, stayed well within predefined acceptance criteria throughout.
  • Linearity and Range: Does the ELISA response scale predictably with concentration? The assay demonstrated strong linearity across the quality control panel, consistent with predefined acceptance criteria, confirming the working range established during pre-validation.

Customer Outcome

  • A validated cGMP anti-inflammatory functional assay with documented specificity, accuracy, precision, and linearity
  • A detection ELISA range carried forward from pre-validation and confirmed during formal validation
  • Reproducible inhibition results across three operators and three days, with all nine validation runs meeting every acceptance criterion
  • Signed final pre-validation and validation reports

Client Impact

The client now has a validated functional assay for measuring the anti-inflammatory activity of its cell therapy drug product, backed by a documented path from early assay characterization through formal cGMP validation.
